Pakistan politician Imran Khan has 3 fractured vertebrae

LAHORE, Pakistan (AP) ? Prominent Pakistani politician Imran Khan is expected to make a full recovery despite fracturing three vertebrae and a rib in a dramatic fall at a political rally, his doctor said Wednesday.

Khan, a former cricket legend whose party is a leading contender in this week's national elections, toppled about 5 meters (15 feet) off a forklift Tuesday that was raising him and three guards to stage at a rally in the eastern city of Lahore. The dramatic footage was broadcast repeatedly on local TV.

The 60-year-old Khan fractured a vertebra in his neck and two in his back in the fall, said his doctor, Faisal Sultan. He also cracked a rib and cut his head, but Sultan refuted earlier reports that Khan fractured his skull.

"The most important thing out of all this is the spinal canal is intact and Mr. Imran Khan is in full control of all his limbs and his body functions," Sultan said at a press conference in Lahore. "We are very confident that all these fractures will heal with time and will heal completely and allow him to be fully, completely functional and fit as he always is."

Khan will be kept on bed rest in the hospital for at least a few more days so doctors can conduct further examinations, Sultan said. Two of the guards who feel with Khan have been treated for their injuries and discharged, while the third is still in the hospital and is scheduled to have an operation on his leg, Sultan said.

Khan's injuries will largely knock him out of the last few days of campaigning before the elections are held on May 11, although it's unclear whether he will try to address any political rallies by phone or video.

It's possible that Khan could benefit from the accident if Pakistanis choose to vote for him out of sympathy.

Hours after the fall, the charismatic politician gave an interview from his hospital bed. He was visibly shaken and had a cut on his forehead, but he was still asking people to vote for his party.

"I have done whatever I could do," Khan said. "Now you have to decide whether you want to make a new Pakistan."

One of Khan's main competitors, former Prime Minister Nawaz Sharif, suspended his campaign for a day on Wednesday out of respect for his opponent.

The fall put a damper on what has been one of Pakistan's most dynamic election campaigns. Khan earned legendary status in the country when he led the underdog national team to a 1992 cricket World Cup victory, and had injected new energy into a political system long dominated by dynasties.

He entered politics in the late 1990s but it wasn't until 2011 that his party, Pakistan Tehreek-e-Insaf, gained a widespread national following. Now his party is considered one of the three main challengers in the upcoming election.

Many analysts consider former Sharif's Pakistan Muslim League-N party to be the front runner in the election, but Khan has emerged as a wildcard. His party has dented the two-party system long dominated by the Pakistan Muslim League-N and the outgoing Pakistan People's Party and could steal votes from both.

Much of Khan's support has come from young, middle-class Pakistanis in the country's major cities, a potentially influential group. Almost half of Pakistan's more than 80 million registered voters are under the age of 35.

According to a recent poll by the Pew Research Center, 60 percent of respondents viewed Khan favorably. However, that figure dropped slightly from a year ago, and now Khan is slightly outranked by Sharif. The Pew survey, which polled 1,201 people, was conducted in March and has a margin of error of plus or minus 4.3 percent.

Khan had been setting a furious pace of rallies and election events across the country to drum up support for his campaign. Few expect him to be the next prime minster, but his party could play a role as a kingmaker or form a solid opposition in parliament.

The election will mark a historic transfer of power from one democratically elected government fulfilling its full term to another, something that has never happened in Pakistan's coup-checkered history.

But the vote has been marred by near-daily violence.

A suicide bomber detonated a car packed with explosives outside a police station in the city of Bannu in northwest Khyber Pakhtunkhwa province Wednesday, killing a policeman and two women, said police officer Rafiq Khan.

A bomb hidden in a vegetable cart exploded in a market in another northwest city, Hangu, wounding 14 people, police officer Azmat Khan said.

Protein complex may play role in preventing many forms of cancer

May 5, 2013 ? Researchers at the Stanford University School of Medicine have identified a group of proteins that are mutated in about one-fifth of all human cancers. The finding suggests that the proteins, which are members of a protein complex that affects how DNA is packaged in cells, work to suppress the development of tumors in many types of tissues.

The broad reach of the effect of mutations in the complex, called BAF, rivals that of another well-known tumor suppressor called p53. It also furthers a growing notion that these so-called chromatin-regulatory complexes may function as much more than mere cellular housekeepers.

"Although we knew that this complex was likely to play a role in preventing cancer, we didn't realize how extensive it would be," said postdoctoral scholar Cigall Kadoch, PhD. "It's often been thought that these complexes play supportive, maintenance-like roles in the cell. But this is really changing now."

Kadoch shares lead authorship of the study with postdoctoral scholar Diana Hargreaves, PhD. Gerald Crabtree, MD, professor of developmental biology and of pathology, is the senior author of the study, published online May 5 in Nature Genetics.

Chromatin-regulatory complexes work to keep DNA tightly condensed, while also granting temporary access to certain portions for replication or to allow the expression of genes necessary for the growth or function of the cell.

Members of Crabtree's laboratory have been interested in BAF complexes and their function for many years. Recently, they reported in the journal Nature that switching subunits within these complexes can convert human fibroblasts to neurons, which points to their instructive role in development and, possibly, cancer.

"Somehow these chromatin-regulatory complexes manage to compress nearly two yards of DNA into a nucleus about one one-thousandth the size of a pinhead," said Crabtree, who is also a member of the Stanford Cancer Institute and a Howard Hughes Medical Institute investigator. "And they do this without compromising the ability of the DNA to be replicated and selectively expressed in different tissues -- all without tangling. In 1994 we reported that complexes of this type were likely to be tumor suppressors. Here we show that they are mutated in nearly 20 percent of all human malignancies thus far examined."

The researchers combined biochemical experiments with the data mining of 44 pre-existing studies to come to their conclusions, which would not have been possible without the advent of highly accurate, genome-wide DNA sequencing of individual human tumor samples. Interestingly, mutations to certain subunits, or particular combinations of mutations in the complex's many subunits, seem to herald the development of specific types of cancer -- favoring the development of ovarian versus colon cancer, for example.

The importance of the BAF complex as a tumor suppressor is further emphasized by the fact that, in some cases, a mutation in one subunit is sufficient to initiate cancer development.

"For example," said Kadoch, "a type of mutation called a chromosomal translocation in the gene encoding one of these newly identified subunits, SS18, is known to be the hallmark of a cancer called synovial sarcoma. It is clearly the driving oncogenic event and very often the sole genomic abnormality in these cancers." Kadoch and Crabtree published a study in March in Cell uncovering the mechanism and functional consequences of BAF complex perturbation in synovial sarcoma.

The startling prevalence of mutations in the BAF complex was discovered when Kadoch conducted a series of experiments to determine exactly which proteins in the cell were true subunits of the complex. (The exact protein composition of the large complex varies among cell types and species.) Kadoch used an antibody that recognized one core component to purify intact BAF complexes in various cell types, including embryonic stem cells and skin, nerve and other cells. She then analyzed the various proteins isolated by the technique.

Using this method, Kadoch identified seven proteins previously unknown to be BAF components. She and Hargreaves then turned to previously published studies in which the DNA from a variety of human tumors had been sequenced to determine how frequently any of the members of the complex were mutated.

The results, once the newly discovered members were included, were surprising: 19.6 percent of all human tumors displayed a mutation in at least one of the complex's subunits. In addition, for some types of cancers (such as synovial sarcoma), every individual tumor sample examined had a mutation in a BAF subunit. The results suggest that the BAF complex, when unmutated, plays an important protective role against the development of cancer in many different tissues.

The researchers are now focused on learning how the mutations affect the tumor-suppressing activity of the BAF complex.

"We certainly want to further our understanding of the mechanism behind these findings," said Hargreaves. "Do they promote cancer development by inhibiting the proper progression of the cell cycle? Or perhaps they affect how the complex is positioned on the DNA. We'd like to determine how to recapitulate some of these mutations experimentally to see what types of defects they introduce into the complex."

Other Stanford authors of the study include postdoctoral scholar Courtney Hodges, PhD, and former lab members Laura Elias, PhD, and Lena Ho, PhD.

Rolling Stones launch tour with energetic set

(AP) ? The Rolling Stones must have passed through a time machine before taking the Staples Center stage to kick off their "50 and Counting" anniversary tour.

Mick Jagger, Keith Richards, Ronnie Wood and Charlie Watts delivered a rousing set Friday night with the kind of youthful energy and musical verve they displayed decades ago. Their faces showed their age, but their performance did not, with a nonstop, more than two-hour set packed with well-worn hits.

Jagger and Richards are each 69 years old, and Watts is 71, but they transformed into their younger selves for the night. (Wood is the baby of the band at 65.)

Jagger might not hit all the notes he once did, but he still busted out his almost spastic, serpentine dance moves on song after song. He's impossibly thin, and his spine showed through the light shirt he wore.

Staples Center was packed to capacity for the concert, the first of 17 dates the Stones are set to play throughout the United States.

The stage was modeled after the band's iconic logo, with lips and teeth above the stage and a tongue-shaped platform that extended into the crowd.

A video of famous folks sharing their favorite Rolling Stones memories played before the band took the stage, with Johnny Depp, Martin Scorsese, Perry Farrell and others reminiscing about their favorite tunes.

Actress Cate Blanchett recalled "just how skinny they were."

"It really, really pisses me off," she said.

(Honestly, the Stones could produce a best-selling diet book ? the members are as slim as they were as aspiring rockers in the 1960s.)

Other videos showed the aging rockers as young men.

Jack Nicholson was among the stars in the audience, and fans welcomed him with a round of applause as he took his seat.

"It was either us or the Lakers, so now you got us," Jagger said early in the show, referring to the basketball playoffs that forced the band to postpone its opening concert from Thursday to Friday.

"It doesn't matter to Jack Nicholson," Jagger continued, "because he was coming to both of them."

Nicholson wasn't the only star in the house. Gwen Stefani, wearing long blond hair and a bedazzled Rolling Stones tank top, joined the group onstage to sing "Wild Horses."

"I've got to get one of these T-shirts," Jagger said, admiring her top.

Keith Urban played guitar and sang backup on "Respectable." Former Stones member Mick Taylor added guitar to "Midnight Rambler."

Richards sang a pair of tunes: "Happy" and "Before They Make Me Run." Jagger also played guitar and harmonica, and came out in a floor-length marabou cape to perform "Sympathy for the Devil."

"We first played in LA in 1965," Jagger said. "Thank you for keeping on coming to see us."

The Rolling Stones' tour continues through June 21.

Tesla CEO amends Model S financing deal after criticism

DETROIT (Reuters) - Tesla Motors Inc, the automaker led by billionaire Elon Musk, overhauled financing strategy for the Model S electric sedan in response to criticism that its initial plan last month overstated the potential cost savings.

Changes include allowing buyers 72 months, instead of 63, to repay loans, lowering monthly costs by more than one-fourth. Tesla also guaranteed that the resale value of the Model S would top other luxury cars in its segment.

"We looked at it and decided we were wrong," Chief Executive Musk told reporters during a conference call on Friday. "Other people were right, and we needed to fix it."

The financing deal is intended to make the Model S electric sedan, which starts around $70,000 before a $7,500 tax credit, more affordable.

But Tesla got flack from critics who said the company was too aggressive in saying payments could be as low as $500 a month. The company's online payment calculator valued time saved filling up on gas at $100 an hour.

The automaker also drew criticism for pegging the resale value of the Model S to the Mercedes S class of sedans.

Tesla has since changed the default settings of its online payment calculators to be more conservative. The calculator now reflects only potential gas savings and assumes premium gas will be below $5 a gallon over the next three years.

Tesla will also guarantee that the resale value of the Model S will be higher than comparable luxury vehicles from BMW , Audi , Mercedes , Lexus or Jaguar , the company said.

"We're guaranteeing that the resale value in three years will be better than that of any of the major premium sedan brands," Musk told reporters.

Without taking into account gas savings, the monthly cost of owning a Model S will be around $800, down from $1,100, Musk said during the call. Including gas savings, the cost will be between $500 and $600 a month.

If 70 percent of the miles put on the car are for business purposes, the depreciation benefits cuts the "effective monthly cost" of owning a Model S down to $350, Tesla said.

The new financing deal will be extended to those who financed their Model S purchases over the last month. At least one-fifth of buyers over the last month made use of the financing deal, but Musk told reporters that he expects that figure to go as high as 70 percent.

Acer Aspire R7 Hands On: We're Not Quite Ready for This Kind of Crazy

Can we start by saying the R7 is nuts, and that that's totally great? Because it is, it's great that Acer's doing this weird new thing that doesn't exactly make sense, just to see if it works.

The very first thing you notice about the R7 becomes at once one of the the most and least important things about using it: the Ezel hinge. It's very godo at some of its jobs, and not very good at others. The problem is that it's got more to dot han the typical hinge. It's got to not only open and close like a typical laptop, but adjust forward and backward like an actual easel. Angling the screen with the top part of the hinge feels perfect. Tilting and flipping it all the way around to the other side (a novelty, but one you might actually use) takes just the right amount of force. But while you'll appreciate the back/bottom part of the hinge being so rigid while you're using the R7 as a touchscreen monitor?there's no wobble at all?it makes opening and closing the lid, and further, adjusting it forward or backwards, pretty inconvenient. That sounds like a small detail, and plenty of people won't mind a clumsy, two-handed process adjusting the hinge, but it's something you'll be doing quite often, given the trackpad placement.

About that. I'm not sure it makes that much ergonomic sense. The unit I used was on a raised box?about the level of a standing desk?and I didn't get to sit down with it on my lap. But I got the sense that the preferred use method is going to be with the screen mostly covering up the trackpad. That's where you'll see some benefit from the design, but it's still questionable, given standard placement would allow your hands to still be near the screen, but still have access tot he trackpad. And on the lap, where the wrist rests can also act as a stabilizing mechanism, this makes even less sense. There's definitely a type of user who's going to love this, though.

The most disappointing thing about the R7, though, is that despite its naming lineage, it seems like a big step backward from the S7 in build quality. It's sturdy, to be sure, but its plasticy and uncomfortable keyboard are a step back from the S7's, which was polarizing as far as usability, but always felt and looked premium. The 1080p 15-inch screen is very pretty, though large enough to be goofy when folded all the way down in tablet mode. It's not overly heavy, for a 15-inch device, but it ain't light, either.

The R7 is pretty clearly aimed at people who will be using it at a desk all day, and who have some compelling reason to be using touch over other input methods. It doesn't really make much sense for anyone else. For now, that first group is small enough to not register on a wide scale. That might not be the case in the future, but for now, the R7 really isn't for everyone.

NEW HYDE PARK, NY A recent study by pediatricians from the Cohen Children's Medical Center of New York examined to what extent pediatric physicians adhere to American Academy of Pediatrics (AAP) clinical guidelines regarding pharmacotherapy in treating young patients with Attention Deficit Hyperactivity Disorder (ADHD). The results showed that more than 90 percent of medical specialists who diagnose and manage ADHD in preschoolers do not follow treatment guidelines recently published by the AAP.

"It is unclear why so many physicians who specialize in the management of ADHD -- child neurologists, psychiatrists and developmental pediatricians -- fail to comply with recently published treatment guidelines," said Andrew Adesman, MD, senior investigator and chief of developmental and behavioral pediatrics at Cohen Children's Medical Center in New Hyde Park. "With the AAP now extending its diagnosis and treatment guidelines down to preschoolers, it is likely that more young children will be diagnosed with ADHD even before entering kindergarten. Primary care physicians and pediatric specialists should recommend behavior therapy as the first line treatment."

Current clinical guidelines for pediatricians and child psychiatrists associated with the American Academy of Child & Adolescent Psychiatry (AACAP) recommend that behavior therapy be the initial treatment approach for preschoolers with ADHD, and that treatment with medication should only be pursued when counseling in behavior management is not successful.

The study also found that more than one-in-five specialists who diagnose and manage ADHD in preschoolers recommend pharmacotherapy as a first-line treatment alone or in conjunction with behavior therapy. Although the AAP recommends that pediatricians prescribe methylphenidate when medication is indicated, more than one-third of specialists who prescribe medication for preschool ADHD said they 'often' or 'very often' choose a medication other than methylphenidate initially (19.4 percent amphetamines; 18.9 percent non-stimulants).

"Although the AAP's new ADHD guidelines were developed for primary care pediatricians, it is clear that many medical subspecialists who care for young children with ADHD fail to follow recently published guidelines," said Jaeah Chung, MD, the study's principal investigator who also practices at Cohen Children's. "At a time when there are public and professional concerns about over-medication of young children with ADHD, it seems that many medical specialists are recommending medication as part of their initial treatment plan for these children."

Citrix Launches Podio Chat To Bring Instant Messages, Video And Audio Chat To Its Social Business Platform

logo-podio-blue-CMYKCitrix today added a new real-time chat feature to its Podio social business platform that allows users to discuss projects, tasks, documents and other items by using standard instant messages, as well as audio and video chats. Citrix argues that this makes Podio, which was already being used by over 200,000 companies at the end of last year, “the first collaboration service of its kind to provide IM and video chat ?in context.? While Podio, which Citrix acquired last year, is quite a fully featured business platform and already included a number of real-time features, including real-time comments, the new chat feature allows for one-on-one and group chats. It features all of the usual chat features you’ve likely seen on Yammer or tools like Convo, including the ability to like comments. Citrix notes that Podio Chat is unique in its ability to bring text, audio and video chat together in one platform. Because Podio also allows you to invite users from outside of the company (contractors, suppliers, partners, etc.), the new chat feature should also make it easier and faster to work with them. As for the video chat tool, Citrix says that it will allow for ad-hoc, face-to-face discussion, as well as allow you to set up recurring team video meetings using the company’s GoToMeeting service. This feature, Citrix says, will allow businesses to unify their communications on its Podio platform “by enabling teams to communicate in a variety of different ways to get work done, all without email or third-party communication tools.” The new chat features will be available on the desktop, where it will live in a new chat pane, as well as on Podio’s mobile apps for Android and iOS.

Newtown officials discuss plans for Sandy Hook school

NEWTOWN, Conn. (AP) ? Newtown, Conn., officials are meeting to discuss what to do with the elementary school where 20 first-graders and six educators were shot to death in December, but they might not vote.

A consultant for the town said a task force of 28 local elected officials likely wouldn't make a decision Friday night and will return for another meeting next week.

Residents have expressed mixed opinions on what should be done with Sandy Hook Elementary School.

The task force has narrowed a list of choices to renovating or rebuilding on the school site or building a new school on property down the street.

The 430 surviving students are attending a renovated school renamed Sandy Hook Elementary School in the neighboring town of Monroe.

This 3D-Printed Cyborg Ear Comes With a Built-In Antenna

(AP) ? Authorities have ordered evacuations of a neighborhood and a university about 50 miles west of Los Angeles where a wildfire is raging close to subdivisions.

The blaze on the fringes of Camarillo and Thousand Oaks broke out Thursday morning and was quickly spread by gusty Santa Ana winds. Evacuation orders include California State University, Channel Islands.

About 100 miles to the east, four residences have burned in a grass fire that has prompted the evacuation of an elementary school in Jurupa Valley.

Other fires are burning around the state with various degrees of containment.

Mo. GOPs at odds over what to do with extra money

JEFFERSON CITY, Mo. (AP) -- After coping through several lean years, Missouri's Republican legislative leaders now are at odds over whether to save or spend an apparent excess of money.

Senate Appropriations Committee Chairman Kurt Schaefer says he expects Missouri will end its fiscal year on June 30 with about $400 million more than had been projected. He wants to spend more than $200 million of that to replace an aging state mental hospital in Fulton.

But House Budget Committee Chairman Rick Stream, R-Kirkwood, prefers to save the money as cushion for next year's budget while waiting to see if the economy continues to improve.

The difference of opinion among Missouri's top legislative budget writers will have to be resolved soon. Lawmakers face a constitutional deadline of May 12 to send a budget to Gov. Jay Nixon for the fiscal year that starts July 1.

Schaefer, R-Columbia, has been pressing House members to revise the revenue projections agreed in December ? upon which the budget is based ? because tax revenues have been better than expected. But Stream and other House leaders have refused to do so.

"Being momentarily ahead of the consensus revenue estimate is not a good enough excuse to me to engage in a new large amount of additional spending," House Speaker Tim Jones, R-Eureka, told reporters Monday night. "I just don't see that that's fiscally prudent, fiscally conservative or anything the House has done in the past."

Schaefer contends House members are acting as if the excess revenues do not even exist.

"It's some kind of alternate reality in this building where generally accepted accounting principles don't seem to apply," Schaefer said.

Missourians should get an update soon about state revenues. Tuesday was the final day of April, and monthly revenue figures typically are released a few days after the close of the month.

Linda Luebbering, the budget director for Democratic Gov. Jay Nixon, has said previously that revenues are running ahead of projections, but that may diminish in coming months.

Although AMD hasn't had much recent success at the higher end of the processor spectrum, it's clear the company is very much interested in attacking the budget realm. Witness its two new lower-tiered FX-series processors: the quad-core FX-4350 and six-core FX-6350 bring noticeable (if modest) improvements over the FX-4300 and FX-6300 chips they effectively replace. Both unlocked CPUs up the base clock speed to 3.9GHz, while the Max Turbo speeds jump to a respective 4.3GHz and 4.2GHz each. The FX-4350 further trumps its ancestor by doubling the Level 3 cache to 8MB, albeit with a higher power draw. More importantly, prices aren't budging much at all in spite of the brisker performance. When the FX-4350 and FX-6350 cost just $122 and $132 each, they're cheap enough that DIY builders shouldn't weep when the bills are due.
